报告摘要:Cell-penetrating peptides (CPPs) have gained great attention since they are promising bio-tool to enable delivery of various molecules in non-invasive manner.   With this method, unlike methods that use viruses, there are no side effects such as carcinogenesis from DNA damage since protein is penetrate directly into the cells via physiological process.   Actually, conventional CPPs such as TAT and polyarginine are extensively utilized in numerous studies of non-invasive molecular delivery, however, they non-selectively permeate broad range of cells including non-neoplastic and neoplastic cells.  We created novel artificial CPPs that are selectively and efficiently incorporated into human tumor cells according to their lineage.  Tumor lineage-homing CPPs were isolated from a random peptide library generated by mRNA display technology.  Their advantageous tumor cell-targeting ability was corroborated in an in vivo mouse model for imaging metastatic human tumors, and by employing CPP-fusion anti-tumor peptide in vitro and in vivo to induce tumor growth inhibition.  The CPPs would be available to various medical fields such as diagnostic imaging technology, molecular therapeutics, by utilizing these as a tumor-specific carrier for proteins, nucleic acids and chemical compounds.  Thinking these possibilities, we hope our study will provide a clue to solve one of the unsettled issues in oncology, covering a broad medical science in future.
