MOLECULAR MECHANISMS OF CYTOKINE GENE EXPRESSION AND THEIR IMMUNOLOGICAL ACTIVITIES IN INFLAMMATORY DISEASES

Interleukin-12 (IL-12) plays a critical role in the activation and function of T lymphocytes during microbial infection. IL-10 has potent anti-inflammatory and immunosuppressive activities on leukocytes. The production of IL-12 and IL-10 by antigen-presenting cells is regulated in opposite manners during innate and adaptive immune responses.

Understanding the molecular mechanisms controlling the gene expression of IL-12 and IL-10 in interactions between pathogens and the immune system is essential to efforts to develop therapeutic strategies for infectious, malignant and autoimmune diseases.
We are currently investigating in the following areas:

I. Molecular mechanism of the regulation of IL-23 gene expression during phagocytosis of apoptotic cells by macrophages and dendritic cells, and its impact on the development of Th17 cells in autoimmune lupus.

II. Molecular mechanisms of the regulation of IL-12 gene expression in APCs by the Crohn’s disease-associated mutants of nucleotide-binding oligomerization domain 2 (Nod2) and IL-12-mediated inflammatory bowel disease.

III. Molecular and cellular mechanisms whereby cancer cell-derived novel molecules inhibits dendritic cell-induced T cell activation and T cell-mediated antitumor immunity.

IV. Role and mechanism of CCL5 in the development of myeloid-derived suppressor cells in triple negative breast cancer.

Her research focused on exploring new enzymes for industrial application, probing the enzyme structure-function relationship, and studying the molecular evolution of enzymes. In recent years, besides stability mechanism of thermophilic enzymes, regulation mechanism of catalytic activity and enzyme molecular evolution, our research focus gradually shifts to redesign the microbial key metabolic enzyme and to reprogram the drug synthesis pathway based on the concept of synthetic biology. This contains exploring the structure-function relationships for the key enzymes in the biosynthetic pathway of drugs, redesigning these enzymes by protein engineering technology, and then improving the non-natural substrate selectivity, catalytic efficiency and adaptability to the industrial environment, at last achieving the optimization and artificial reconstruction of drug synthesis route.

Brief Introduction of Research Direction

Prof. Li’s major research interest is marine microbial symbionts in sponges and corals. The marine biosphere is well-endowed with symbioses. As one of the oldest multicellular animals, i.e. more than 600 million years old, marine sponges (phylum Porifera) probably represent one of the most complex symbioses on the earth. Corals (more than 500 million years old) are called as the rainforests of the oceans, which also harbor abundant and diverse microbial symbionts. Marine microbial symbionts are suggested to play an important role in the host’s chemical defense, nutrients and energy transfer as well as the marine C/N/P cycle, and are important resources for marine natural products and functional genes. The present research includes:

1. Marine natural products and enzymes from marine microbial symbionts, biosynthesis and fermentation.

2. Community structure and functions of marine microbial symbionts, the association of sponge holobiont/coral holobiont.

3. Omics of marine microbial symbionts.

4. Response and adaption of sponge holobiont and coral holobiont to environment and climate changes.

Brief Introduction of Research Direction

1. Bioprocess engineering and molecular biochemical engineering: Our work focuses on medicinal mushroom fermentation and plant cell cultures for production of useful secondary metabolites, such as regulation of biosynthetic processes of antitumor ganoderic acids and ginsenosides.

2. Industrial biotechnology and advanced fermentation technology: Our lab has been doing platform building for efficient antibiotics expression by microbial systems, such as study on validamycin A and ansamitocin fermentation, including effects of environmental factors on antibiotics fermentation process at molecular levels of genes and enzymes, and improvement of the biosynthetic pathway via metabolic engineering.

3. Bioenergy and environmental biotechnology: We are interested in the low-cost efficient production of ethanol and other chemicals using lignocellulosic hydrolysates, production of biohydrogen by synthetic biology, and the role of quorum sensor in biodegradation of toxic aromatics.

Main research direction:

By combining in vivo and in vitro electrophysiological, calcium imaging, and animals’ behavioral approaches, we focuses on the cellular mechanisms of synaptic plasticity and learning & memory, the pathogenesis and early intervention of epilepsy and depression, and effects & underlying mechanisms of acupuncture. He has published 23 SCI papers with a combined impact factor of 150 and over 760 citations. His research has been supported by grants from National Natural Science Foundation of China, the Shanghai Committee of Science and Technology, as well as ‘973’ program grant, Shanghai Pujiang program. He serves as the Executive Committee Member of the Shanghai Society for Neuroscience and the Shanghai Society of Biophysics.

Brief Introduction of Research Direction

Microbial ecology of complex communities, such as in wastewater treatment system, oil reservoirs and soil environment.

1. By using the cutting edge molecular biology techniques, we compare samples from industrial wastewater treatment system in different operational status and in different scale, both in industrial scale and in lab scale bioreactors. Not only the diversity of microorganisms in such environment is extensively investigated, but also the main members which are responsible for pollutant degradation are most concern for our research.

2. Denitrifying bacteria are the most interested target group in our research among others. The bacteria and functional genes related denitrifying degradation were identified and associated with the denitrification efficiency of bioreactor and soil environment.

3. We intensively compared the microbial community structures of water injection wells and their adjacent production wells in the oil reservoir. And we also constructed anaerobic crude oil degrading consortia which showed distinct degradation of crude oil both in sulfate reduction condition and methanogenic condition. Aim of these studies is trying to understand the mechanism and regulation of anaerobic transformation of the unreachable crude oil in subsurface oil reservoirs to methane.
