摘要:Virtually all biological phenomena are of dynamic nature, though the absolute kinetics of different processes can vary drastically. This fact has motivated the development and application of new mass spectrometry and bioinformatics tools for the study of biological processes in my group during recent years. The presentation will first cover the comprehensive analysis of adaptation of Corynebacterium glutamicum to a) osmotic stress and b) heat stress with an emphasis on the recently developed versatile protein turnover technology in a gel-free proteomics workflow, which is advantageous for membrane proteins. Thereupon, it will be discussed how the daunting complexity of protein modifications can be addressed in an untargeted proteomics approach. Own developments in this area will be illustrated by an application study of oxidative stress response. The final topic will be the development of highly accurate and sensitive MS-based protein and metabolite quantification methods to develop kinetic models of metabolic pathways. These methods enabled to identify new lipid modifying enzymes in the endoplasmic reticulum of the sphingolipid-producing yeast Wickerhamomyces ciferrii and to model and interpret metabolic differences between different strains.
